Compared with the traditional low-pressure gas discharge plasma source, atmospheric pressure room temperature plasma (ARTP) has the characteristics of low plasma jet temperature, uniform discharge and high concentration of chemically active particles. Based on ARTP technology, Our company, together with the Department of chemical engineering and the Department of Engineering Physics of Tsinghua University, has jointly developed the first special instrument for mutation and breeding of microorganisms by means of plasma — ARTP Mutagenesis Breeding Machine. The instrument has high mutation rate, compact structure, simple operation, high safety and fast mutation speed. A large mutation library can be obtained by one mutation operation (within a few minutes), which greatly improves the intensity of strain mutation and the capacity of mutation library; ARTP technology combined with high-throughput screening technology can realize rapid and efficient evolutionary breeding of organisms.

Case1 : Application ARTP Streptomyces Avermitilis to Incresae Production of Avermectin 

Avermectin is a substance with anti insect activity produced by Streptomyces avermitis fermentation. It has high application value in medicine, agriculture and animal husbandry production with high efficiency and low toxicity. At present, the production strains of Avermectin in China have the problems of low fermentation unit and high production cost. It is the key to adopt new mutation technology to breed avermectin high-yield strains.
When ARTP was treated for 3 minutes, the mortality rate of Streptomyces avermitis was as high as 98.2%, the mutation rate and positive mutation rate were 30% and 21% respectively, and the yield of avermectin increased by 23%.

Case2 : Application of ARTP to Mutate Escherichia Coli to Increase Production of Threonine 


Threonine is an important essential amino acid, which is widely used in medical, food and feed industries. The production methods mainly include protein hydrolysis, chemical synthesis and biological fermentation. It has become the preferred method for threonine production to improve the performance of strains by mutagenesis and then carry out biological fermentation.
When ARTP was treated for 1.5min, a high-yield strain of threonine was obtained, and the acid production in shake flask reached 50.6 g / L, which was increased by 99.6% compared with the original strain. After 50 passages, the heredity was stable.
